Proper selection of solvents and column chemistries. If water is running and one tries to use silica column, it is big issue. Filtration of samples. Washing of column using proper solvent sequence are some the areas where one can make a mistake

I am looking for an HPLC which is fast, sensitive detection system and capable of hyphenating with other instruments. I would like to have an HPLC system with column chemistry capable of separating different hydrocarbon classes (saturates,cyclic saturates, unsaturates, aromatics in petroleum distillates ( As an analyzer).

One should understand the configuration of HPLC being asked for and analysis types to be performed on the machine. Similarly election of column chemistries also play important role in the success of any HPLC system. It is always good to use universal and selective detector in combination (e.g UV and RI detector) for any method development activities.
